A System for Measuring the Melting Temperature of DNA

The goal of this project is to build a system that can measure the melting temperature of double stranded DNA.
The system will be incorporated into the Advanced Bio-engineering Laboratory.

This project consists of the following parts:

  1. Building and verifying the electronics, first on a solder less breadboard, and then on a printed circuit prototyping board.
  2. Once the electronics are verified, we would like to then design a custom printed circuit board (using tools such a KiCAD or PCB Express), and have such a board manufactured.
  3. Improve the LabView control program for this system. This includes adding an auto zero function, converting the control loop to a finite state machine implementation, and implementing the system under Linux.
  4. Using the system to measure DNA melting behavior. The system will be test on DNA solutions of 80bp, 40bp, and 20bp fully matched strands, 20bp 50% mismatched strands, and 20 bp fully mismatched (i.e. different random) strands.
